My idea:
-1. nix-instantiate could generate the derivations of all evaluable expressions
-2. the build-dependency directed graph would be extracted from these
in some suitable format
-3. standard graph reachability algorithms could be used

I hope I understood you correctly.

For example, I tested ffmpeg by simply grepping ffmpeg and building
the affected packages :-)
